I do not think that there are any exact methods to calculate the amount of deposited polymer other than doing a full chemical analysis (and even that will be difficult)

You could get the weight af the deposited layer but this will include unknown amounts. Depositing the polymer on a quartz crystal microbalance (taking the softness af the deposited layer into account) will give you the mass of the deposited polymer plus additional contributions from solvent and electrolyte salt incorporated into the conducting polymer layer during deposition. If you are able to get the weight the electrode before and after careful removal of solvent by drying, you will still have an unknown amount af electrolyte salt, and the ratio between salt and solvent in the film will probably be different from the ratio in the bulk electrolyte due to Donnan exclusion.

If you have a really clean system without any parasitic side reactions you may use the synthesis charge, and subtract the charge of the first reduction of the polymer to the neutral state (compensating for the "doping" of the polymer occurring simultaneously with  chain growth). Assuming that the polymer chains are infinitely long, you can then calculate the amount og monomer units knowing that addition af one monomer unit requires two electrons - but as no one knowns the actual length of the polymer chains, you will even in this ideal case get a (small?) error.

I agree with Dr. West about the various contributions to the total mass of an electrodeposited conducting polymer layer.

The question is how big an error one may tolerate or accept for a reasonable analysis. In our previous study of electro-deposition of polypyrrole using an EQCM, fairly good agreement between theoretical prediction and experimental results were obtained. PS: I was assuming you synthesize under galvanostatic conditions - under CV conditions even very small parasitic currents will offset your results substantially!!

I synthesized self doped polymer galvanostatically. And how can be the charge of the first reduction of the polymer?

 If you know your polymer well, you may have identified the potential below which the polymer is reduced (CV is good for this type of analysis). After synthesis you can then reduce your polymer (either potentiostatically or galvanostatically) to that potential and count the charge.

If it is a thick film, probably it can be obtained by weighing the substrate with and without film using a microbalance.
