Hi, is it fine to use protein refolded in LDAO or SB-12 detergent for thermal stability experiment with CD spectroscopy? CD spectra as a function of temperature. Will LDAO or SB-12 have influence (false results) on spectra with higher temperatures or is it safe to use it like this? I want to check thermodynamics of protein unfolding (and thermal stability in general)
Usually when you do CD with temperature gradient you keep the wavelength fix at 222 nm for proteins with a high content of a-helices, or 218 nm for proteins with a high content of b-structure and detergent should not interfere with signal at these wavelength. You can easily check that by doing CD spectra of a blank with LDAO or SB-12 and run the same temperature gradient you will run for your protein. You should have a flat line or a very low signal noisy line.
I was worried that detergent will get little turbid or will change unspecifically the signal with higher temps. You are right I will check the detergent alone.
