I am getting spores in the form of clumps. I have tried making the suspension with different concentrations of tween 20 but I am not getting proper spores. What is the correct protocol for harvesting spores from a submerged culture.
Deeksha the clumps are the mycelium (it's obvious), use cheesecloth for filtering the mycelium from the suspension, and you can see the spores in the supernatant (liquid medium).
Usually, you will have a very high number of spores, so dilute it accordingly and use it for fermentation.
This suggestion maybe a bit OTT for your purposes but density gradient centrifugation would work well to separate the denser spores from the mycelium. Just be sure to agitate vigorously before separation so spores detach from mycelium. Ludox or Percol work well for gradient making, there maybe other suitable alternatives for gradient medium. Viveks suggestion should also work if you use a mesh with pore sizes just slightly larger than the spore itself.
It would be better if you can highlight whether the objective is to harvest the spores or you just want to separate. If you need spores it's best to do it through SSF or just in agar plates. As it can be easily removed by scraping from the surface in case of agar media or can be harvested using 0.1% tween with shaking so that spores get suspended in the liquid.
