We just got a Bead Mill 24 homogenizer, does anyone know the cycles and time needed to crush frozen plant leaves? We are using 2ml tubes with 2.4mm crushing beads.
Thank you, to everyone who followed, read, and provided information on this question. A special thanks to J.C. Tarafdar who provided excellent guides. After a few attempts we have developed a technique that works with our plant tissue samples.
Using 2ml screw cap micro centrifuge tubes with 8 2.4mm metal beads 1. Using 50mg of leaf tissue frozen in liquid nitrogen cut these leaves with a razor or break up the leaf tissue.
2. Add the broken leaf tissue to the 2ml screw cap micro centrifuge tube
3. Using the OMEGA E.Z.N.A. SP Plant DNA Kit add Sp1 buffer and RNase to the micro centrifuge tube.
4. Place tubes in bead mill.
5. Using the following settings: Speed: Highest, Time: 45 seconds, Cycle: 2, Delay time: 30 sec, Temp: 25 C.
Using this method we were able to obtain a highly concentrated DNA extraction.
