I need HL-60 as neutrophilic models as the cell line is near to diploid and I need to edit by CRISPR/CAS9.
I tried to differentiate HL-60 with 1uM ATRA for 5-6 days but I reached only 5-10% of monocytes instead of neutrophils.
By using 1,25% DMSO I had no differentiation.
Does anyone knows other strong differentiating agents?
Hi, from our experiences the major issue is the source of the cells. We tried with two different sources, what worked really great for us were the cells purchased from Sigma (98070106). The cells differentiated well after ATRA, DMSO or DMF treatment.
Hi, HL-60 cells are very sensitive. When I differentiated them I used retinoic acid (1 uM or 10 uM) for 24h. I couldn't see any particular difference between the two concentrations. I hope you'll manage :) Good luck!

thank you for your experience... Unfortunately ATRA for me produced only rare (5-10%) monocytes. I have tested 1uM and 5uM with no differences...
I will try as suggested by Aneta with DMF, otherwise I think thath my batch may have some difficulties in differentiating
