I am going to be growing the three mention strains and just wondering if trypticase soy yeast extract medium be ok for all three strains or do i need to grow them on more selective media ?
I recommend to retrieve the liophillized strains in BHI broth. TSB might be a good alternative too. One the broth cultures are grown, I would suggest Columbia Blood Agar. This protocol should work well.
Thank you. Could you suggest how long I should leave them grow in the broth before streaking them on the plate ? How would you go about building up stocks of the strains .ie freezing them ? Sorry for all the questions its my first time working with Liophillized strains .
Thank you so much for your help. Just a few more things, should i do an OD before i plate them or just plate them after 24hrs ? Also should I test the them on selective agar just to make sure the bacteria is the strain i want ?
It always help to check OD at 550 or 600. Please check that it is higher than 0,6-0,8. This would mean that it is sufficiently grown as to culture it in agar plates.
If you use Columbia Blood Agar it would be OK. But you can check the purity of the culture by taking a drop of the broth culture and observe it under the microscopy. In addition, once the plate cultures are grown check that you only have one type of colony in the plate.
