I already have 30 sample. I want to also know if we want to analyze the gene polymorphism in rare diseases and we don't have enough sample which other methods rather than case/control study should be used.
You definitely want to do a case-parents trio study. Here is a review, which demonstrates that the association power is stronger in case-parent trios than case-control studies when disease prevalence is rare.
as Jan said, searching for analyzing rare disease with so poor prevalence is not suitable with cases/controls study. Familial studies are best, trio is a major way to manage genetics background correction. You need now to recruit the families and why not in case of large families run linkage analysis.
