I am currently doing my Final Year Project to synthesis ester using Lipase from Rhizomucor mehei that had been immobilized on eggshell via adsorption. After washing it with buffer I stored it in desiccator to minimize water content before dry it back to powder form.
Should I store it in refrigerator or is it okay to store it in desiccator before I could dry it using freeze dry ?
Assay the residual lipase activity in dessicated as well as refrigerated immobilized preparations over a period of time. Say every week. You can seek answers to your questions by performing this experiment only.
Amir, did you check the residual activity of egg shell particles-bound lipase?
As mentioned by Sir Shamsher, I advise you to test the activity of your enzyme before and after immobilization but also before and after washing. This measure must be done in the same conditions, namely in a "standardized manner"
When testing the activity as already recommend, keep in mind that enzyme activity is generally strongly influenced by the water activity. Normally you would keep a salt in your exicator in order to have a specific water activity, different possibilities are available. The same should be done with your solvent (assuming you are using an organic solvent for your esterification).
You must be aware that water (aw) acts as a lubricant for a lipase. Therefore, if you perform a too intense (or long) dissection you may disrupt the activity of the enzyme and thus reduce the effectiveness of your application.
