How long are you leaving the cells to adhere before changing media?

How long is the biofilm growing?

I know people who have had good results from the CV method in this paper PMCID: PMC165379 (based on this method PMCID: PMC123944). Quite straightforward, just be careful to wash and rinse gently.

Actually we are leaving the cells for 18h  in media and 24h  for biofilm formation. I have tried washing by pipette, since its hard and taking long time. Hence, I am searching for another method. I know some people who had result more than OD 1.5 or 2. But I am not getting such results when i rinse using tub.   

Have you looked at the paper by Nett et al. 2011? They used XTT but its worth a look.

Antiadhesive activity of the biosurfactant pseudofactin II secreted by the Arctic bacterium Pseudomonas fluorescens BD5.

The lipopeptides pseudofactin II and surfactin effectively decrease Candida albicans adhesion and hydrophobicity.

It worked really great for us.

As for washing step, we used PBS buffer and multichannel pipette to wash wells really gently.

I will refer those papers. But it is really hard to do if we increase number of plates at the time. I wonder how people using pipette without disturbing the cells. Thank you for response once again.

May I know what kind of medium that you used to culture the biofilm?  Media can be one of the factor that lead to the low amount of biofilm.  I have published a paper which has been accepted in late April this year.  Here is the title 'Polymicrobial biofilm formation by Candida albicans, Actinomyces naeslundii and Streptococcus mutans is Candida albicans strain and medium dependent'.  Arzmi et al. 2016. Medical Mycology.  This paper explained in a very detail how the morphology of the C. albicans affect the biomass of biofilm in 96 well plates.  Hope this helps.

Actually we are using PDB for this experiment and facing problem at the time of washing the cells.
