i have a purified compound from plant source, it is hydrophobic in nature. it readily dissolve in DMSO, Ethyl acetate and methanol. Once i add water it will get precipitated, i want to check this compound with SF-9 cell line.
you can try Cremophor EL or polysorbate 20, non-ionic solubilizers and emulsifying agents used for parenteral administration of various poorly-water soluble drugs. You can also use cosolvent (by mixing water, ethanol and PEG 400 for example) this can improve the solubility of your compound, another approache consists in using complexation with safe cyclodextrins such as HPBCD or SBE7BCD, you will increase the solubility and the drug will be delivered to cells by quick release from complex .
hi sir, thank you for your reply i tried with polysorbate 20 but still little precipitation was there and will not it cause any negative effect on cell lines sir?
Maximum tolerated DMSO in cell culture is 1% and 0.1% is safe. From your stock solution in DSMO dilute your compound at the desired concentration in 1% polysorbate 20 (or polysorbate 80) to obtain the working solution to be added to the cells. For controls use identical concentrations of DMSO and polysorbate.
