I have varied the glyphosate concentration to derivatize with 20 nM of FMOC-Cl but i am not getting linearity for the derivatives for my calibration curve. I used UHPLC-ToF-MS/S, negative polarity for my analysis .
you have to use a surplus of FMOC-Cl to get a quantitative conversion. Otherwise you have the effect that your calibration function shows no linear course. Excess FMOC-Cl can be intercepted with aminoadamantane.
