By just subtraction and addition of plasma  you can get the desired HCT of blood sample. 

Is there a protocol such as centrifugation speed and other details, please?

  Practically, we are doing i.e take the anticoagulant whole blood in two tubes from the same patient. Centrifuge these tubes at 2000 rpm for 3 min at room temperature to get the clear plasma on the top. Than add or substrate plasma for one tube to other to get the desired HCT of 40% - 60%.If you know the HCT of original sample,  You can easily get the desired HCT by manipulating the volume of plasma.
