I´m trying to make a plaque assay of NDV on DF1 cells. Infected for an hour, and then put some DMEM (no FBS) 0.8% methylcellulose with 10% allantoic fluid. But it doesn´t seem to solidify. Any suggestions!?

Use agarose 0.8% for overlaying, put at room temparture for 1hour. And then put plate in incubator. Good luck !

Hi Carlos...
IAfter 1 h of incubation at 37 °C, inocula should be discarded and monolayers should be covered with MEM with 1% methylcellulose, or 0.8% in your case... remember it does not solidifies it is just like a gum or something like that!
what I generally do is to prepare 2x DMEM and 2X MC and then mix them toghether and incubate with that.
