Ad: Whatman FTA cards are used for biological sample collection, storage, transport, and isolation of high-quality DNA for downstream applications. In the following studies, FTA cards were evaluated for their ability to collect, store, and isolate high-quality RNA from a variety of crude biological samples.
Yes. I have tested a few. Here is a protocol:
(Use of Norgen Biotek Total RNA Micro Kit)
1. Cut out 3 x 3 mm (1/8 inch) diameter circles from a dried blood spot with a single-hole paper puncher and place in a clean microcentrifuge tube.
2. Add 300 μL of the Lysis Solution and vortex for 10 seconds.
3. Incubate the sample at 42°C for 30 minutes. Apply vortex for 15 seconds after every 10 minutes.
4. Briefly spin the tube to collect any drops of liquid from the inside of the lid.
5. Transfer the supernatant to a new microcentrifuge tube. Measure the volume.
6. Add 1 volume of 70% ethanol to the sample (i.e. 100 μL of 70% ethanol for every 100 μL of lysate) and mix well by vortexing for 10 seconds.
7. Proceed to “Section 2: Total RNA Purification from All Types of Lysate” in the product insert of Norgen’s Total RNA Purification Micro Kit (#35300).
The yield you could get is ~ 100 ng per punch-out (it could vary from person to person however). The RNA integrity is of course not as good as anything fresh. However, I tested some mRNA at 4 - 7 day after collection and they were still detected (surprisingly with almost the same Ct as Day 0). microRNA is also tested and again had no issue which is expected (miRNA tends to be quite stable).
