We should be able to use the Qiagen PowerFecal HT kit for 96 extraction of samples without the QiaCube robot, as the spin column/S-block assembly is the same as for the Plant 96 kit. However, Qiagen only provide buffer volumes up to the point of loading on the robot... from checking the low-throughput kit, they add 1200ul C4 to 750ul of lysed DNA. The HT kit at this point loads 320ul DNA (if using proteinase K) onto the robot and the next step is addition of C4, so I assume 512ul (maintaining same ratio) will work, but has anyone tried this and can confirm. Note the LT kit does have some differences ie buffer C2 is not used in the HT kit so the volumes appear different even though starting with ~80% of the fecal volume.
