We are interested in studying the expression of mRNAs in different cell types. The idea is to localize the expression of mRNAs with specific cell types in the tissue itself.
but I have been getting a fair amount of literature for a protocol called Stelaris. Basically it is a type of FISH to detect where your mRNA is in a cell.
Short version of how it works is you have a ton (60 or more) of short probes per target what each have the same fluorophore. so, each mRNA strand will bind multiple probes increasing the signal. It might be good for FFPE due to FFPE chewing up and spitting out RNA.
