I am interested in examining the status of autophagy in peripheral monocytes from patients with neurodegenerative disorders such as Alzheimer's. We take around 5ml of blood from patients and are trying to examine autophagy using a dye that stains autophagosomes. First issue we are facing is that we do not get enough monocytes from PBMCs isolated from 5ml blood. We are taking the adhering population. We were able to acquire only 2000 events. Any comments or suggestions?
