Hi I am going to test some peptides as positive control for my ELISA but don't know what sort of concentration to start of with. Any suggestion will be very much appreciated.
This will be empirical and you will need to do a titration. For positive controls a good target is in the 0.6-1 OD range. You could probably start at 1 ug/ml peptide concentration and serially two-fold dilute, selecting the concentration that provides your optimal OD.
he positive control concentration for a peptide ELISA (Enzyme-Linked Immunosorbent Assay) can vary depending on the specific assay, the peptide being tested, and the overall sensitivity of the assay. The positive control is used to ensure that the ELISA is working correctly and to provide a reference point for interpreting the results.
Manufacturer's Recommendations: If you're using a commercially available peptide ELISA kit, the manufacturer might provide recommendations for the positive control concentration. This can be a valuable guideline.
