Has anyone tried to immobilize hAChE to CM5 Biacore Chip using Biacore 3000?

The sensorgram produced from the immobilization showing very strange behaviour, after activation with EDC/NHS, and upon injection of hAChE, the response begins to be negative, and then very slowly increase..

Regardless the activity of the enzyme, the immobilization of the enzyme should be the same in behavior. Maybe I was not clear, when I am talking about the response, I mean the reponse during injection of the enzyme itself during immobilization, thanks.
