I know this is a strange question, but I accidentally made a stock for Western Blot (TBST and BSA) with an antibody that cannot be used for WB. And someone in my lab said that as long as TBST doesn't burst the cells, its worth trying the FACS analysis with the TBST-based antibody. So yesterday I took some cells and seeded it in 96 well plates and I incubated one with PBS and one with TBST (0.1%) for an hour at 4C (as one would do in a regular sample prep for FACS) and after an hour I looked at the microscope and the cells didn't seem to have bursted. I am wondering, though, if TBST would cause any other problems like compatibility with the FACS machine or anything else. Should I try, for curiosity sake, running the analysis with TBST or is it too risky do you think?
