I'm using oxygen microelectrodes and have some nice oxygen concentration profiles orthogonal to the root surface at different positions. Oxygen concentration profiles seem to vary quite a bit from the tip to the base of the root hair. I also did some some time series at defined positions to record oxygen increases at the root surface.
Both data, time series and oxygen profiles, can, theoretically, be used to calculate potential diffusion rates but I’m not sure what the best data set is and what I have to consider for this calculations. I'm using freshly cut roots immersed in agar solution (1 %) to prevent (minimize) diffusion from the atmosphere. Any advice?
