We did a couple of trials with intact pGWB600 vectors as well as linearised pGWB600 vectors (at the XhoI site). We did overnight incubation for the LR reaction; still, we do not get any colonies. We are confident that the LR protocol is fine because we get successful LR cloning with other vectors. If you have any tips on troubleshooting this, please let me know.
