Has anyone experienced complications when trying to LR clone using pGWB600 series vectors (around 12kb) as the destination vectors?

We did a couple of trials with intact pGWB600 vectors as well as linearised pGWB600 vectors (at the XhoI site). We did overnight incubation for the LR reaction; still, we do not get any colonies....

Has anyone worked with pVYNE(R)/pVYCE(R) BiFC vectors to detect protein-protein interactions between nuclear localising proteins?

The two vectors pVYNE(R) and pVYCE(R) seem to be prone to produce false positives in BiFC. I would like to know whether someone out there has done some work with these vectors for proteins that...
