I would like to have one for my RP open columns. Please share your experience and thoughts. Also please share what kind of pump you have employed for your work.
I've used peristaltic pumps and pumps from SSI. I used columns from Ace Glass (one of a few vendors) because they have fittings that allow easy attachment of pump tubing.
For a gradient, I used two flasks (beakers work too), each with a "jag" on the bottom. The jags were connected by flexible tubing and the tubing was closed with a clamp. The flask I would pump from would be filled with a weak solvent and placed on a magnetic stirrer with a magnet; the other flask was filled with the same volume of strong solvent. I would start the stirrer and the pump and remove the clamp. As the "weak" solvent was pumped, the strong solvent would drain from the other flask and be mixed. This would create a gradient from weak to strong solvent.
It's not as nice as an automated flash system but it worked well enough.
Yes is a pretty cheap and easy way to increse flow and reduce time. But be carefull, too much pressure can break the column, and assure that column never run dry
Yes you can use a pressure pump like aquarium air pump, it will give sufficient pressure to run a normal cumulus. To use this type of pressure pump, you need to connect the pump with a column by using "Zero bed" connected with tubing at the top of the column.
