Hello Prateek,

Can you please tell me if you are not getting insulin expression in control as well as treatment or only treatment?

I have performed an experiment with insulin ELISA kit, where I could see insulin secretion in RIN5f cells at 5.5 mM, 11.1. mM and 25 mM (hyperglycemic condition) glucose concentrations. Although I have not performed mRNA expression for insulin, I believe since the insulin secretion is glucose dependent process, changing glucose concentration might help. You may also confirm it with insulin ELISA method. If you are getting positive results for ELISA, you might need to change your RTPCR parameters.

It was part of your post which states that you are not getting optimum insulin expression. An obvious question is what do you call optimum!
