I actually perform protein extraction from pancreas, which is to me one of the most sensitive organs, in terms of degradation by proteases.

Right after sacrifice, put your (cut) duodenum in a microtube that you will throw in liquid nitrogen.

Then I use precellys tissue homogenizer. Organs are transfered to pre-filled beads tubes, recovered in lysis buffer, and processed in the precellys. Do this on dry ice !

Note that your RIPA must contain 2 fold the usual protease/phosphatase inhibitors.

You will certainly have to work on the buffer volume since these organs contain a lot or proteins, so you might have too concentrated samples.
