I am veterinary pathologist and I am working on Pneumocystis in pigs. We want to establish a method and would need unstained slides from Pneumocystis jirovecii positive AND negative humans. Most publications deal with PCR or cytology, therefore it is really difficult to find somebody who may have formalin fixed paraffin embedded material. I really hope that there is any possibility to get material by this way. Thank you!

Obtaining human material might be quite tricky due to legislation restrictions.
I will suggest that you search for a commercial vendor of such material like Asterand.
Other option is to use PAS (Grocott) positive slides from your collection of pig samples and to establish the assays. As a negative control one can use also a pig sample with definite pneumocystis negativity.
By the way, what is the method which you would like to establish: IHC, in situ hybridization or PCR on slide?
I can dig out one mouse sample with pneumocystis but it is quite limited focal infestation.

I have already found somebody who will co-operate with us. I want to test a human Pneumocystis antibody solution by IHC. Aim is to use this solution for immuno-precipitation for Pneumocystis in pig BALF. The initial steps are the establishment of a functional IHC on human samples and on pig samples.

In that respect: will you perform the precipitation on lysates or on native lavage? There will be difference between lysed sample and organ sample in terms of protein conformation. Lysis leads to primary protein structure and therefore there can be difference in the epitopes.

we will use the native samples. A method has been described for human Pneumocystis and we will try to adapt the protocol.
