Has any one used CD11b / c antibodies for long-term (3-6 months) blocking/depletion of monocytes in mouse?

I'm not sure this is an appropriate strategy. CD11b will hit monocytes, neutrophils, some B cells. CD11c will be relatively useless in terms of infiltrating monocytes. If I were you, I might consider instead ccr2. See http://www.ncbi.nlm.nih.gov/pubmed/15516494 for a bit more. ccr2 and ly6c are generally expressed on infiltrating and inflammatory monocytes but Ly6c is also found on T cells, so not a good choice.

Thanks both : Noel ; I suggest CD11b for 3 reasons

1) the inf. monocytes in this tissue are CD11b/CD11c/Cd45hi/Lys6 +

2) we have already used a CD11b KO mouse to show that this is protective; however we dont have time to to backcross a double knockout into our strain in time for grant renewals

3) our microarray studies show CD11b, CD11c to be candidate genes

Please don't regard any of the following questions/comments as patronizing:

Did you mean Ly6G or Ly6c? If Ly6G then you might be looking at neutrophils. But these would be CD11c-. So, I assume you meant Ly6c. If so, are they just positive or high?

If they are high, then CCR2 would be a great choice. Your CCR2+ cells would include the CD11b cells you wish to target but leave alone other CD11b+ cells, specifically neutrophils. I am not certain whether alveolar macs express CCR2, but I am pretty sure not. They are CD11b- and CD11c+. In essence, it would be a much more targeted manipulation than CD11b which would blow the #$%^ out of a huge number of cells, thus obscuring your rationale for claiming infiltrating macs. If you tag tissue-resident cells (myeloid DC for example) expressing CD11b, you might inadvertently clear those as well via complement mediated mechanisms. Then, what can you really say about the monocytes? You can't because you also affected another large subset of key APC. And so on...
