This issue relates to how insects work and how they are studied, and it has connections to molecular ecology and biochemical analysis. Microlepidopteran larvae have a soft outer layer and a small internal body volume, so standard puncture and centrifugation methods, used in studies on Drosophila, can lead to gut rupture, melanisation and clotting, which damage the samples and skew the results. A gentle method therefore must (i) minimise tissue trauma, (ii) immediately chelate divalent cations and lower pH to suppress phenoloxidase‐driven melanisation and coagulation, and (iii) deliver pooled volumes sufficient for DNA extraction while preserving enzymatic activity when required. These requirements are consistent with classic findings that haemocyte degranulation initiates coagulation and melanisation at wounds, and with reviews emphasising the central role of haemocytes in cellular and humoral immunity.¹–³

Biochemical control of clotting and melanisation.

Two complementary anticoagulant systems are widely validated in insects. First, the low-pH EDTA/citrate “Mead buffer” (≈98 mM NaOH, 186 mM NaCl, 17 mM Na₂EDTA, 41 mM citric acid, pH 4.5) arrests clotting, stabilises haemocytes for separation on Percoll, and is now routine from lepidopterans to mosquitoes.⁴–⁷ Second, a mildly acidic trisodium-citrate/citric acid buffer (100 mM/17.8 mM, pH 5.5) preserves phenoloxidase (PO) activity for functional assays; addition of phenylthiourea (PTU) prevents melanin formation when activity is not being quantified.⁸ This logic follows broader mechanistic work showing that coagulation localises immune effectors and restricts pathogen spread, so prompt chemical suppression is essential when the analytical endpoint is nucleic acid integrity or unbiased cell counts.¹,²,⁹

Osmotic considerations for small larvae.

Cell integrity depends on matching anticoagulant osmolality to haemolymph. Recent refinement of Drosophila larval haemolymph collection estimated true osmolality near 306 mOsm kg⁻¹ under melanin-free conditions; large variations among published salines were traced to contamination and melanisation.¹⁰ Although microlepidopteran values are not yet resolved, targeting ~300 mOsm kg⁻¹ and working on ice reduces lysis and preserves analytes; the standard Mead buffer is hypotonic (~160–370 mOsm reported depending on preparation), which supports rapid chelation and PO suppression but argues for minimising residence time before downstream dilution or processing.⁴,¹⁰

Collection strategy optimised for microlepidoptera.

For soft, small larvae, a superficial micro-incision with ophthalmic scissors at the head capsule margin yields cleaner haemolymph than lateral body wall puncture, reducing fat body and gut contamination. In honeybee larvae, head incisions combined with immediate capture of exuding haemolymph produced the lowest turbidity and highest purity among five tested tools; this principle transfers to small larvae where intestinal rupture is a major risk.¹¹ For tiny coleopterans, a gentle “tear” at leg bases with subsequent cold centrifugation through a perforated tube yielded clear plasma; this provides an alternative when head access is impractical.⁸ For wild caterpillars, JoVE protocols emphasise surface sterilisation, chilling, and immediate anticoagulant contact to maintain haemocyte viability—procedures directly applicable to microlepidoptera.¹²

Standard operating procedure (SOP) — gentle pooled haemolymph collection for DNA work, with options for cell or enzyme assays.

Larvae are surface-sterilised in 70% ethanol, rinsed in sterile water, and anaesthetised on ice. Individual 0.5- or 1.5-mL tubes are pre-loaded with 20–50 µL of ice-cold anticoagulant. For DNA extraction and haemocyte counting, use Mead buffer (98 mM NaOH, 186 mM NaCl, 17 mM Na₂EDTA, 41 mM citric acid, pH 4.5).⁴–⁷ For PO activity assays, use 20 µL citrate buffer (100 mM trisodium citrate, 17.8 mM citric acid, pH 5.5); for immunodetection or general protein work where melanisation must be blocked, add solid PTU to the collection tube (e.g., 2 mg per 1.5-mL tube, as validated for Tribolium plasma).⁸ Under a stereomicroscope, a fine ophthalmic scissor tip makes a 100–200 µm incision at the fronto-lateral head margin, avoiding gut tissues. Exuding haemolymph is immediately wicked into a pulled glass capillary or P10 tip and expelled into the anticoagulant. For very small larvae, two rapid, minimal strokes often suffice; if flow slows, a second micro-nick can be placed contralaterally. Samples remain on ice, are gently mixed by inversion to distribute anticoagulant, and the first visibly turbid or discoloured drop is discarded if gut leakage is suspected. Pooled volumes are built from multiple larvae until 1–5 µL haemolymph equivalent is reached, which is typically necessary for robust DNA work. Clear plasma can be obtained by a brief spin (e.g., 10,000–12,000 × g, 5–10 min, 4 °C) to pellet cells and debris, as used for small beetles; for cell analyses, pelleting is avoided and samples are processed immediately.⁴,⁸,¹¹,¹³

Expected yields and pooling rationale.

Yield per small insect individual is typically in the tens to a few hundreds of nanolitres. High-throughput Drosophila adult sampling commonly retrieves ~50–150 nL per fly, while Tribolium methods reported ~5 µL plasma from 30 individuals (~0.17 µL each).⁸,¹⁴ Consequently, microlepidopteran larvae of comparable or smaller mass will often require pooling from 10–30 specimens to secure the ≥1–5 µL haemolymph used in Chelex-based DNA workflows. This expectation is consistent with long-standing population-genetic practice that exploits very small, non-destructive haemolymph volumes when feasible.¹³–¹⁵

DNA extraction compatible with minute samples.

For rapid genotyping, heat the pooled aliquot in 5% (w/v) Chelex-100 suspension, optionally with proteinase K, then use the supernatant directly for PCR. The canonical Chelex protocol uses 5% resin, protease digestion at ~56 °C, and a brief 95–100 °C denaturation before centrifugation; Gerken et al. successfully amplified microsatellites from 1–5 µL insect haemolymph with this approach, and methodological compendia recommend using 2–5 µL of the Chelex supernatant per 10-µL PCR.¹³,¹⁶–¹⁹ Chelex chelates metal ions that catalyse DNA degradation during heating, reduces heme-like inhibitors, and avoids organic solvents, making it well suited to haemolymph.¹⁶,²⁰

Quality control and contamination checks.

Purity can be monitored by turbidity: in Apis mellifera larvae, an NTU window of ~22,432–24,505 indicated low contamination; although absolute values will differ with instrument and species, maintaining consistent low turbidity across replicates is a practical criterion.¹¹ For PO assays, citrate buffer preserved activity and supported reliable detection of zymogen and active forms; Tabunoki et al. detailed dopamine/MOPS/CaCl₂ kinetics suitable for small plasma volumes.⁸ If haemocyte work is envisaged, immediate anticoagulation, low temperature, and pH control are critical to prevent degranulation and clotting artifacts; classical and modern studies document how rapidly these processes proceed after wounding.¹–³,⁹

Ethical and survival considerations.

Haemolymph withdrawal can be non-lethal in larger insects, enabling longitudinal designs, but in very small, soft-bodied larvae survival declines with increased blood loss and wounding. Pilot tests should therefore quantify post-sampling survival and growth at the intended sampling volume; when survival is required, limit to a single micro-nick, strictly cap volume, and use head-capsule incisions to avoid gut trauma.¹³,¹¹

A micro-incision, head-capsule collection into ice-cold low-pH EDTA/citrate anticoagulant, supplemented with PTU when melanisation must be blocked, provides a gentle and efficient route to haemolymph from microlepidopteran larvae. Pooling across individuals to achieve microlitre-scale volumes, followed by 5% Chelex extraction, yields DNA compatible with standard PCR genotyping. When enzymatic activities or haemocyte phenotypes are targets, pH, osmolality, and processing time should be tuned using the cited parameters to minimise artefacts while retaining biological signal.⁴,⁸,¹⁰–¹⁵
