I submit an article in the topic of crude oil biodegradation by selected bacterial strains. The editor demand me to determine the growth rate, lag phase and biomass from OD growth curves of bacteria cultured in mineral salt medium supplement with crude oil. How i can do it, since i have only (04) kinetic growth points (0, 3 rd, 7 th and 13 th day)?

I do not know how your growth curve looks like but only four points are extremely hard to get/predict any distinguish phase of bacterial cutlure. I would suggest to explain the reviewer that this experiment focuses mainly biodegradation efficiency over the incubation period. In that case bacterial growth pattern is negligible; how much oil is bedgraded is the important. Anyway, if you would have good bacterial growth curve (maybe typical log, stationary and death phase), you can simply get growth rate by selecting two points data divided by the time taken. You will take Ln of the data (common formula you can find in any microbiology book/google). Biomass from OD, you need to run a calibration curve (probably new experiment with similar condition) of OD versus biomass. Then place you OD of the experiment (already submitted one) and get the calculated biomass.

Please start a new batch experiment once you have inoculated you media collect sample and read optical density. From the starting point OD does not change say about 2-4 hours; then suddenly growth pick up and OD sharply increase as your organism propagates. Please define the length of time when OD did not change; that is your lag phase. NExt step duration of log or exponential phase. I think you can do it one day and overnight the whole experiment get growth curve for any specific organism in a defined media. Wish you good luck
