I am working with Streptococcus equi subspecies equi and co-cultivate those with immune cells. To know more about the growth I do currently the following growth experiment:

All incubation is carried out at 37 °C, 5 % CO2 and with slightly opened lid.

4047 (wt) is grown overnight in THB, centrifuged and resuspended in Tyrode's buffer. A volume according to an end OD of 0.05 is added to 30 ml falcon tubes, three in total. One sterile control is present.

The used medium is 89 % DMEM, 10 % heat-inactivated FBS and 1 % (2 mM) L-glutamine (the same which I use for co-cultivation).

After 5 h the growth stops at an OD of 0.15. Medium after 5 h started to turn orange.

I did not find any literature about growth of equi or other Streptococci in DMEM media. Anyone suggestions why the growth stops?
