Ginkgotoxin is easily dissolved in water or any low molecular alcohol.- The publication of Liu et al gives a misleading discussion of our contributions to the ginkgotoxin problem: 1. we isolated ginkgotoxin preparatively from Ginkgo leaves and Ginkgo medications and identified the products by H1 and C13 NMR and by HPLC- comparison with an authentic sample. Thus, their analysis is fine but does not really constitute much progress. 2. Wada (5) and Jones(4) were not the scientists who showed that Ginkgotoxin is a pseudosubstrate of the pyridoxalkinase-but compare U. Kästner in FEBS Journal 274 (2007) 1036-1045.
The literature has not been presented conscientiously by Liu et al. EL.
Dr Leistner is correct in identifying Kastner et al (2007) as the first identification of ginkgotoxin as a pseudosubstrate for mammalian pyridoxal kinase. Our study by Jones et al extended these observations to the parasite causing sleeping sickness and provided evidence of essentiality and possible selective druggability of the parasite enzyme. The publication by Kastner et al was cited in our article. I trust that our presentation of the scientific literature is beyond reproach!
Attempts to develop a suitable HTS assay for our Drug Discovery Unit proved problematic and a fragment based screen failed to identify suitable chemical start points. We have abandoned work in this area, but anyone wanting reagents to test their good ideas for for drug development are welcome to contact me.
