I intend to isolate a specific gene from Sinorhizobium fredii known to harbor number of plasmid. The sequence of the gene of interest viewed from GenBank originated from a plasmid. The plasmid might be a large on and have no kit available for its purification. So I wonder if the same gene can be isolated from the genomic DNA.
Dear Didier if you only want that particular gene..and If you have specific primers for your gene of interest you could go for Colony PCR and you could isolate your gene of interest from bacteria... all the best
Dear Didier, you can use alkaline Lysis Method for the isolation of Plasmd from Bacteria (http://www.bio-protocol.org/wenzhang.aspx?id=30). or as Prakash suggested, if you know the sequence of your gene why not to design primers and try to amplify it.
