Hi, I'm going to study the effect of cadmium on rat sperm quality and I use the following protocol for mobility/sperm count (without dilution): epididymal spermatozoa were obtained by mincing the epididymis with anatomical scissors in 5mL of physiological saline and incubated at 32 ◦C for 2 min. An aliquot of this solution was placed into a Malessez cells and motile sperm were counted by using microscope at 400× magnification. Non-motile sperm numbers were first determined, followed by counting of total sperm. Sperm motility was expressed as a percent of motile sperm of the total sperm counted
my question is what is the formula for counting the total sperm in this case (without dilution)?

The volume of the whole Malessez chamber (2mm *2,5mm *0,2mm) is 1 microliter. If you count all sperm in the whole chamber field (n), you obtain the sperm number in 1 microliter, the total volume(V) of your sample was 5 ml. The formula for the concentration(c) will be c = n * 1000 [spermatozoa per mililitre], for the total count (t) it is t = n*1000*V e.g. t = n*5000.
