I have prepared the water in oil (W/O) microemulsion of a therapeutic agent. Now to determine the particle size and the poly dispersity index (PDI) of the prepared microemulsion using Malvern zeta sizer, how should I prepare the sample. I have used capryol 90 as oil phase.
I need to know the dilution medium for water in oil microemulsion, Not measured yet..
As per the F. Bordi, you can also try the same concentrated preparations for determining the particle size distribution. We do dilution to eliminate the possibility of aggregated particle which may give interfering results. So, first you try the same concentrated preparation. if you are not getting the proper results then you have to dilute it with external medium. While determining the PDI you should change the reference RI of the medium. Hope it is clear now.
You can take measurements in the same oil phase as such or after making dilution. Just change solvent in setting and select your oil (phase). If it is not present, you will have to add some information manually such as density, refractive index etc of oil.
Please BE CAREFUL TO SELECT OIL PHASE IF YOU ARE USING DISPOSABLE CELLS to carry samples because some organic solvents (if taken as oil phase or to make dilution) react with polymeric cells (cuvette) and will produce misleading results.
