I am currently work to remove fat from a whey protein concentrate with a fat content of ~10 % w/w in order to obtain a defatted powder. I used the folch method but instead of cloroform/methanol I used Hexane to remove apolar lipid followed by Hexane/isopropanol 3:2 for polar lipid.

After the extraction of fat the powder resulted in fine dust with little flakes (before was clumpy).

To validate the procedure, I analysed the sample with rose-gotlieb method without issues during extraction, but when I distillate the solvent for recovery to obtain only the fat in the flash it formed a coagulation similar to egg-white. It only happen when I applyed fat extraction to the powder, indeed with regular powder with fat no issues were visible during distillation and recovery of solvent.
