Hi
I am looking for a fluorescent dye that is sensitive to the local environment with an excitation wavelength of around 480 nm.
Ideally one were the emission maximum wavelength changes in response to local environment.
I know FITC quenches quite strongly, but its emission maximum remains pretty constant.
Does anyone know of a dye in that spectral range?

What aspect of the environment do you want the dye to respond to? Fluorescein responds to acidic pH with a loss of fluorescence. SYPRO Orange is used in the thermal shift assay because it increases fluorescence when it shifts from a hydrophilic to a hydrophobic environment. Various dyes respond to changes in metal ions, such as Ca2+.
One that is sensitive to the local hydrophobicity. Thank you, I will give it a shot.
