Hi All, I just got a weird image from Beckman Coulter Cytoflex S. So I stain Liver mononuclear cells with APC-NK1.1 and BV605-TCRβ, then I got the following image. The left lower quadrant shows cells accounted for 64.6%, but there are few corresponding dots.
Does anyone know the reason? Your advice would be greatly appreciated.
The upper image is weird, the lower looks normal. And these two image were from the same experiment, with same compensation. Only different wells.
There are two problems i could see. The axis on the plots started with 10^2. And the staining in your normal plot is bit higher than the other. This means some cells are hiding from the display in the dot plot. Try changing the axis scale. Good luck

Please fix your scaling in X & Y - axis. If you are using Log scale , it will be ok or if you are intended to stick with Bi-exponential scale, optimize your R values and ensure better visibility for the graph. If you need further assistance, please feel free to knock me - [email protected]
