Good day,

I am evaluating my plant's flavonoid and phenol content, and I have repeated the methods several times, and each time my flavonoid content has shown to be higher than the phenol content, and I don't think it makes sense. What could be the reason?

These are the methods that I used:

Flavonoid method: "a sample (1.0 mL) of the liquid extract was mixed with 4 mL of Milli-Q water, and 0.3 mL of aqueous NaNO2 solution (5% w/v). Subsequently, 0.3 mL of AlCl3 (10% w/v) and 2 mL of 1 M NaOH solutions were added to the mixture after 5 and 6 min, respectively. The reaction mixture was then incubated at room temperature at approximately 25 °C for 60 min. After the incubation, 200 μL of the mixture was transferred to 96-well plate in triplicate, and the absorbance was measured at 510 nm."

Phenol method: "About 200 μL of each concentration gallic acid was taken in a test tube and added 1.5 mL of 10% Folin-Ciocalteu reagent solution. All the experimental test tubes were kept at room temperature in a dark place for 5 min. Then 1.5 mL of 6% Na2CO3 was added to each test tube, and the mixture was kept in a dark place at room for 2 h. Finally, the absorbance of each concentration of gallic acid was measure by UV-visible spectrophotometer at fixed wavelength 760 nm."

Thank you.
