 I hate to do this, but I need to answer your question with a question.  Are you expecting there to be platelet infiltrates inside the lung tissue?  I was under the assumption that while there are resident megakaryocytes in the lungmicrovasculature, the platelets they may spawn will be released into the microvasculature and eventually end up in the circulation.  Any platelets you find logically then will be the same platelets you see elsewhere in the blood.  Unlike looking for lets say macrophages, which can infiltrate into the tissue, the platelets do not if I remember correctly.  I would be hesitant to digest solid tissue to find a cell type that is not a part of that tissue, rather that may simply just be "passing through".  I think what you will end up with is a large amount of lung cells, but very few, if any, detectable platelets.  Can you share what you are actually trying codetermine in these platelets?  It may be easier to do a ligation at the pulmonary artery and pulmonary veins, take samples of the blood before and after passing through the lung, and analyze the platelets using one of the more reliable whole blood flow cytometric protocols.  We were able to measure platelet activation in whole blood samples as small as 10 µL with good reproducibility.  If you want more info you can reply here, or if your interested in confidentiality when responding to the questions you can email me at [email protected]
