I am interested in your experiences and opinions on "Twist Bioscience". I want to order synthesized genes and they seem to have very competetive prices.
Unfortunately they don't have any testimonials or other user recommendations, so I wonder if anybody used them and can say a few words about it.
I have synthesized genes there multiple times and it was pretty good (you really cannot find their price anywhere else). However, their synthesis technique has some limitation. If you just want to synthesize ORFs then they are probably the cheapest company to use but if your construct contains a lot of regulatory repeats they might not be able to synthesize it as it cannot be codon optimized.
Is there a reason people are using Twist for dsDNA fragments over, Genscript? Eurofins? IDT? I guess they claim half the price/high yield? But their delivery/logistics seems to be not as good as the big boys. My research group has had maybe one instance( out of close to a 100) of a gBlock being a bit troublesome and this was quickly resolved with their QC dept and IDT had my fragment two weeks after we put the order in and for half the price due to their lower QC assessment.
Remember, if ya'll are using these for Gibbson/InFusion cloning, you have(hopefully) including the homology arms as a part of the fragment, you can just simply not PCR amplify and just put in 15-30ngs of your fragment straight in. You just have to be more vigilant in getting a solid sequencing read when you are screening, as "misspelled" fragments are more relatively abundant in the total pool of printed fragments and if your gene is somehow toxic to your carrying strain, transformants that have deletions/insertions causing missense mutations will be selected for.
