I'm setting a good protocol for exosome isolation by cellular conditionate medium with Fixed angle BECKMAN COULTER ultracentrifuge. To date I'm working with polypropylene tubes but I don't like that tube becouse they haven't any cap and it does not allow me to see any pellets.
What is your experience in this particular area?
I use polycarbonate tubes for ultracentrifugation of viruses where I need to see the pellet properly. The tube are transparent and strong. The only problem is to take a good care of the tubes since they are sensitive to acids and alcohols. Good luck
If required centrifugation speed is below 30,000 rpm and one could use a swing bucket rotor (instead of a fixed angel rotor), one could use a Corning glass tube with an adaptor. At higher speed (or g value), the tube/adaptor is prone to break in a fixed angel rotor.
