The easiest check is: Check if you are really injecting the calibration standard into the MS/MS. You might not have the calibration bottle filled or the injecting syringe/route is clogged and there is no standard flowing into the MS/MS.

If you have a small vial/bottle that contains your calibration standard, make sure that it has enough material in it, and it hasn't expired.

Another way to check and see if this is the source of your problem, is to inject a sample of pure compound in methanol, such as caffeine in methanol. Use a proper method that retains caffeine to inject that sample. Now look for the response to caffeine in your MS/MS. If you can get a very sharp peak for caffeine, then you MS/MS is good and the problem of tuning is that you are not injecting the calibration standard. But, if you can't get any peak for caffeine, it means that your MS/MS is down.

In this case, start troubleshooting your MS/MS by the ion source. Check the ion source for dirt/carbon build-up and do a routine cleaning. Also check that all other parameters are set properly, such as ion source temp., MS/MS voltage and collision energy, etc.

Troubleshooting can sometimes be lengthy and frustrating, so you need to be patient and don't mess up.

I tried the detector callibration after uploading the method of perfluorotributyl amin PFTBA (Callibration standard), and it was done succesfully. Before this i was callibrating with default compound and i think it was scaning ions other than PFTBA and this was the reason for availability of insufficient ions. This idea may be wrong but as i understand. if it has other reason the kindly share for learning purpose.
