Hi there, I am hoping there is an easy fox here. A student of mine is using the Lowry method to quantify total protein content in microalgae biomass. In running the assay however, he accidentally prepared his first batch of Lowry reagent D in a much stronger solution of sodium hydroxide than intended, i.e. 1 M not 0.1 M. Used to solubilize the microalgae proteins, I just wonder if this high concentration of alkai will denature the proteins and for example, affect their optical absorbance properties for spectrometer readings? Many thanks for your expert assistance.
