I want to extract intracellular material from a Bacteroidetes strain, but I can't use temperatures above 60°C.
Does anyone have experience in the effectiveness of ultrasound, lysozyme or other methods?
Note to self:
- fill up Bacteroidetes suspension with EtOH to a concentration of 70% EtOH
- let shake in sterile bench at 60°C until EtOH evaporated (20min for 100µL in my case)
- repeat once & let dry completely (1h for 100µL in my case)
- resuspend yellow-white-ish residue (in same medium as the suspension had) and test for sterility in LB (no growth in my case)
