I have been trying to measure the overall cell proliferation in rat brain after learning by EdU staining and also wanted to measure the OPC proliferation and differentiation by using EdU incorporation staining.
I injected EdU @ 50mg/Kg body wait 1 hour before training and sacrifice rats after 3 hrs and 24 hrs followed by staining brain tissues (20 um section) with EdU click-it Alexa 555 kit. However I have not actually been able to pick up any staining at all for the EdU kit.
In the literature there are a some groups that have done EdU staining in vivo by injecting IP before training and get good signal and I'm just following their protocol but I don't understand why I am unable to get it to work. If anyone has any suggestions, advice or ideas that I could try they would be much appreciated. Or if any one have some optimized nice protocol please share with me, that I could try they would be much appreciated.
