I am trying to make my cultured cells have overall weak cell attachment to the plate (but not complete detachment). Would adding EDTA to media weaken cell adhesion? If so, how much?
I am also trying to make another set of cultured cells have stronger cell attachment. Would adding more than 10% of FBS into DMEM media help? (I am aware that FBS contains fibronectin that helps in cell attachment to the plate)
What is your cell type? That is important. Many cells deposit their own matrix material.
If you use microbiological plastic petri dishes less fibroctin or vitronectin will adsorb from the serum. This was shown with using radio-iodinated proteins.this should result in weaker attachment. A glass substrate will also give weaker attachment.
For stronger binding try poly-lysine coating followed by high concentration overcoating with purified fibronectin.
You will not get much of a boost in adhesion over 10% FCS. EGTA would make cells sick, but a low calcium medium, like those used to grow basal keratinocyes might weaken adhesion.
