Hi, i am undergraduate doing my final year project..i am struggling to find the difference between using various extract concentration or only one for calculating the RSA%. I am extracting my sample by dissolving 3 g into 30 mL of 80% methanol. Then i have to conduct DPPH assay but i am confuse reading the articles because there are many version states to dilute at various concentration and some only one..is it acceptable to do only 1 concentration of extract for example i dilute until 1 mg/mL because the dpph solution stock is very limited
Testing multiple extract concentrations in a DPPH assay is ideal for accurate antioxidant activity and IC50 determination. However, using a single well-chosen concentration is acceptable for preliminary results if it falls within the assay’s linear response range.
yes, but you have to establish a calibration curve with a standard antioxidant such as ascorbic acid, trolox or others...

It is complicate to stablish a scientific conclusion by using only one concentration. In order to compare the robusstness of your method and compare the data with those already reported, I would use for sure 3 concentrations in my bioassay. This will help to see a concentration dependant activity and confirm no flaws in the process.
