As you know the quantitation of bilirubin in serum (pale yellow) is based on the formation of a reddish-purple colour, and in my case, the colour of serum is blue-green (biliverdin colour). so when I added the reagent the colour doesn't change to reddish-purple, but after 2 hours changed that is mean all the biliverdin convert to bilirubin.
