Different physiochemical parameters play a very important role in inducing the lipase gene .Availability of the substrate and its concentration is one such factor .Unfavorable condition like lack of nutrients ,increased temperature ,have negative effect on lipase induction. Among the different triglycerides used as a substrate as well as carbon source for the production of lipase by number of researchers .Fatty acid composition and chain length of the triglycerides influence the lipase gene induction . Olive oil served as a best substrate for lipase production .

Yes. Olive oil is the best inducer for lipase production. I would like to know the detailed mechanism of TAG (crude as well as emulsified with a surfactant) uptake and Lipase production by Bacteria.  If any review articles related to this please share.. 

In lipolytic microorganisms, lipase production is usually induced by the products of TAG degradation, i.e., fatty acids, with oleic acid being the best inducer. So, yes the faaty acid composition of TAG can have an effect. Gene expression can be regulated by fatty acid response element (FARE) and/or oleate response element (ORE) in yeast for instance but I am not sure this exists in bacteria. I suggest you have a look at papers from Karl-Erich Jaeger's group.

Types of oils and fatty acids do have effect on lipase production. However, culture condition also affects the production for example  Mg+2 might enhance the production. In the paper by Bouke et al. (Appl. Environ. Microbiol. 2007,73:3838-3844) showed that hexdecane and tween 80 stimulate lipase production by differner mechanisms.

 Although this is an obvious phenomenon in biochemical reactions but the concentration must not go higher than that a threshold. In your project, any positive correlation between concentration of lipid and gene expression of lipase. 

The fatty acid composition of TAG does in no way influence the expression of Lipase gene especially in Bacillus subtilis. One of the simplest of all answer to your question is that you check the Km values along with Vm values of the enzymes on different substrate and you should be able to see the substrate(s) having maximum affinity for  the lipase of your study and you will get the answer by your self.  You will find that some of the substrate molecules act as very good triggers or initiators for the gene to be expressed.

Very good question , Dr. Ravi Mohan: Lets go back to our basic biochemistry lessons, You will recall that TAG is an important feature of Blood Lipids or to some extent Liver lipids. Even there the lipase that is responsible for maintaining a balance of TAG in blood is triggered not by TAG but by a small hormone molecule called glucagon. Now coming back to our Bacillus species, normally the hydrolysis of TAG by lipase is not controlled by any of the 3 fatty acids nor by glycerol. It means that most (if not all) lipases of bacteria especially Gram positive types are triggered by carbohydrates like glucose or maltose, and rightly so as the lipases play a very vital role in transportation accross the cell membrane. the cell membrane of prokaryotes is a multifunctional organelle os a prokaryote cell. Activity of lipases in Bacillus spp is controlled by the functional need rather than mere presence of substrate alone.
